NEW DEVELOPMENTS
OF
CANCERTHERAPY
RESEARCH
The prewlence
of
cancer requires
much work.
fhe
2OO2
Nobel Prize
in
Physiology or Medicine was awarded
to Sydney Brenner, Robert Horvitz and
John Sulston for
their
work on
pro-
grammed cell death
(PCD).
As cancers
occur with suppressed
PCD,
tumor
suppressor, e.9.,
Fas,
Bax, or promoter
genes,
e.9.,
Bcl-2, E1B
or a
"mixed"
gene, e.9.,
c-myc,
are highly
relevant.
ln China,
all-trans
retinoic acid and ar-
senic trioxide
have
been employed suc-
cessfully for the treatment of
a
specific
leukemia (APL), a process related to
PCD.
Arsenic
is
also active against lym-
phatic malignancies and
a
malignancy
ol
plasma
cells
in
the
marrow. Stem
cells are "pleuripotent" cells with
mul-
tiple development potentials; they could
mop up residual cancer cells. Stem cell
lransplants
are also useful for dissemF
nated lupus and Crohn's
disease
(chronic inflammatory disease of the
bowel). Cancer vaccines
are
usually
genes administered to the cancer
pa-
tient
so as to attack cancer cells in a
similarway
as
conventional vaccines
for
infective diseases, usually as
a
preven-
tive
measure
-
adjuvant therapy after
major ablative modalities.
The delivery
of
the antigen
is
now through the incor-
poration into
recombinant
viruses that
carry the
antigen;
alternatively, dendritic
cells are
employed.
There appears to
be
some initial encouraging
results,
however,
further
and more extensive
testing
in
the clinical perspective should
be performed.
